IDTERT 

a multiplex kit for TERT gene promoter mutations detection in tumor DNA

The ready-to-use   IDTERT kit is a multiplexed and sensitive tool for the detection and quantification by digital PCR (dPCR) of TERT (TElomerase Reverse Transcriptase) gene promoter mutations involved in telomere length deregulation.

Telomerase is an RNA-dependent DNA polymerase that maintains chromosome length by adding the telomere, a TG-rich structure, to each end of the chromosome. Mutations of the TERT gene promoter, located at 5p15.33, have been identified in several tumors` types, such as glioblastomas (69%), oligodendrogliomas (68%) and anaplastic oligoastrocytomas (44%). Such mutations are as a new interesting biomarker for the brain tumor classification by molecular biology. These alterations are associated to poor prognosis in many cancers, such as bladder tumors (66%), anaplastic thyroid tumors (42%), cutaneous and basal cell squamous cell carcinomas (59 and 41%).

 Technical specifications

This optimized kit provides a multiplex solution allowing, for each sample, to simultaneously amplify, in one PCR reaction, TERT promoter mutations at position 124 bp and 146 bp upstream of the translation start codon (ATG), corresponding to the  C228T and  C250T mutations (Ref: IDTERT(b)-25), respectively. 

This kit can be used on circulating cell-free DNA (cfDNA) extracted from plasma samples or DNA extracted from FFPE tissues. For all other matrices, please contact us.

It is compatible with the following instrument: Digital Droplet PCR QX200 (Biorad) and Naica System Crystal Digital PCR (Stilla). For other instruments, please contact us.
